Abstract Based on COI barcode sequence, DNA molecular identification was carried out on the
genuine raw materials of Manchu medical deer skin gum, sika deer skin and its adulterants (red deer,
deer and other deer species similar to sika deer are regarded as adulterants), and NJ tree was estab?
lished to analyze the difference between the genuine sika deer skin and the adulterated sika deer
skin, so as to establish a fast and effective identification method of authenticity. The results showed
that in the NJ tree, all the samples of the genuine sika deer skin were clustered into an independent
branch, with a 100% support rate and obvious unilinearity. The adulterated samples were also aggre?
gated into a single branch, with a support rate of 100%. This study proves that DNA barcode technol?
ogy based on COI sequence can effectively identify the genuineness of sika deer skin,which provides
a new scientific method for authenticity identification of sika deer skin, a traditional Manchu medici?
nal material.
